Biomimetic glass nanopores employing aptamer gates responsive to a small moleculew
Single synthetic nanopores are of interest as potential mimics of biological pores such as gated ion channels. Controlled transport through these synthetic pores can be achieved by integrating polymers responsive to chemical or physical stimuli. Stimuli reported to date include pH, ionic strength, temperature, UV light as well as recognition of an ion, a small molecule or a protein. The respons...
متن کاملDisplacement and hybridization reactions in aptamer-functionalized hydrogels for biomimetic protein release and signal transduction.
A variety of hydrogels have been synthesized for controlling the release of signaling molecules in applications such as drug delivery and regenerative medicine. However, it remains challenging to synthesize hydrogels with the ability to control the release of signaling molecules sequentially or periodically under physiological conditions as living cells do in response to the variation of metabo...
متن کاملProtein Detection by Nanopores Equipped with Aptamers
Protein nanopores have been used as stochastic sensors for the detection of analytes that range from small molecules to proteins. In this approach, individual analyte molecules modulate the ionic current flowing through a single nanopore. Here, a new type of stochastic sensor based on an αHL pore modified with an aptamer is described. The aptamer is bound to the pore by hybridization to an olig...
متن کاملRational design of a small molecule-responsive intramer controlling transgene expression in mammalian cells
Aptamers binding proteins or small molecules have been shown to be versatile and powerful building blocks for the construction of artificial genetic switches. In this study, we present a novel aptamer-based construct regulating the Tet Off system in a tetracycline-independent manner thus achieving control of transgene expression. For this purpose, a TetR protein-inhibiting aptamer was engineere...
متن کاملMolecular simulation studies of hydrophobic gating in nanopores and ion channels.
Gating in channels and nanopores plays a key role in regulating flow of ions across membranes. Molecular simulations provide a 'computational microscope' which enables us to examine the physical nature of gating mechanisms at the level of the single channel molecule. Water enclosed within the confines of a nanoscale pore may exhibit unexpected behaviour. In particular, if the molecular surfaces...
